Pertinent and Adaptive Dynamism of the Memory System in the Brain
要旨
The brain maintains proper functions, such as appropriate behaviors or cognitions, despite its lifelong remodeling and perturbation. Memory is a good example of such brain function grounded upon an optimal balance between stability and dynamic changes. For example, long-lasting changes in synaptic efficacy are widely accepted as the biological trace of memory. Past experiments demonstrated that these changes are required for successful acquisition of memory. Recent observations in awake animals, however, revealed high turnover of dendritic spines and the representational drift in the hippocampus, raising the question of whether retention of the experience-dependent changes is necessary for retention of memory. To tackle this fundamental problem, we utilized a recently developed mouse model of artificial hibernation. During the artificial hibernation, hippocampal neurons showed substantial reduction in their activities and extensive elimination of dendritic spines and synapses. Three-dimensional reconstruction of dendrites from electron microscopy images indicated that enlarged spines, believed to support stable memory, were not preferentially spared from spine elimination. Nevertheless, behavioral tests after arousal showed that mice had intact or improved retention of their hippocampal memories acquired prior to hibernation. In the seminar, I will share our recent findings about the network architecture exclusively spared during the remodeling, potentially serving as a structural trace of memory retention. Additionally, I will share another study from the lab, which suggests the existence of a universal homeostatic target that enables the brain to achieve flexibility and robustness for proper functioning.
